The RTP file type is primarily associated with Gromacs.
What is an RTP file?
- Usage: When creating a topology for a protein molecule defined in a "Protein Database" (or PDB) file, GROMACS uses an RTP file to read the interaction type data, as well as an ITP file to read parameter data.
- Structure: e.RTP is a text file that contains defines the default interaction type between the four bonded interactions and residue entries. These interactions are made up of atoms, bonds, angles, dihedrals, and impropers. It is also possible to define different parameters for these residue entries inside the RTP file, which would override the parameters defined in an ITP file.
- Compatibility: GROMACS supports the main operating systems, as well as any other system that has a C/C++ compiler.
How to open RTP files
You need a suitable software like Gromacs to open an RTP file. Without proper software you will receive a Windows message "How do you want to open this file?" or "Windows cannot open this file" or a similar Mac/iPhone/Android alert. If you cannot open your RTP file correctly, try to right-click or long-press the file. Then click "Open with" and choose an application. You can also display a RTP file directly in the browser:. If you know the file format, you can also find out how to open or edit the file. The first characters in a file identify the file format, for example, HTML files start with the bytes <html>. File extension, signature and file format must match; otherwise errors will occur when the file is opened. If there are problems opening a file, you must first determine the file format used. The RTP file extension is used by a few programs, each with its own file format. The file formats most commonly found in RTP files are:
- 40% of all RTP files are RTPatch update/diff data files. These files contain neither text nor words, but only data. Most of these files are 930 bytes to 62 MB in size. The file type was developed 20 years ago and is still in use today. Examples of file names are Patch.rtp. Such files sometimes refer to english, splintercell and emea.
- 6% of all RTP files start with the bytes CD B1 9A E0 74 53 B5 AE, which are crucial for this file format. These are binary files, so they do not contain words or text. The file size is in the range of 290 KB to 990 KB. The following keywords are characteristic: lj%qv.

How to solve problems with RTP files
- Associate the RTP file extension with the correct application.
- Update your software that should actually open residue topology data files. Because only the current version supports the latest RTP file format. Search, therefore, e.g. on the University of Groningen manufacturer website after an available Gromacs update.
